Description
Project Title: Soil molecular probe field sampling for the fungus Albatrellus ellisii or A. fletti. The long term goal of this work is to have a reliable method with which to easily and cost-effectively sample for fungal species from the mycelium in the soil without having to rely on locating the seasonally and annually variable fruiting of fungal fruiting bodies known as sporocarps. This work will be based on FY2004 work in which species specific primers were developed and field tested at known sites for two fungal species Albatrellus ellisii and Albatrellus fletti. The objective of this contract is to determine the relative variability of the mycelium of one of the fungal species, Albatrellus ellisii or Albatrellus fletti, in the soil with: ?X One visit: the relative variability by sampling at several locations where the species is known to occur, ?X Additional visits: the relative seasonal variability by sampling at several locations at different times of the year. In this first effort, soil samples will be collected at three sampling locations where the species is known. The sampling locations will occur over the broad range of habitat types the species occupies in Oregon and Washington. The Contractor shall analyze the collected data for relative proportion of detection estimates (a.k.a. occupancy) at each sampling location and across all locations and test the data for significant differences in proportions per location. An optional additional effort would allow for sampling at the same sample locations at different times of the year to characterize the variability in the relative proportions for seasonal differences.
