Description
NAICS Code: 541380 The US EPA intends to negotiate on a sole source basis, under authority of FAR 6.301, with Battelle Memorial Institute Pacific Northwest Division. This requirement is to develop microarray chips for detecting 6 pathogenic bacteria present in environmental water samples. The chips developed will be validated using pure cultures of bacteria and spiked and unspiked field samples. In 1998 the US EPA listed Aeromonas, Helicobacter pylori and Mycobacterium avium complex (MAC) on the Contaminant Candidate List (CCL) for regulatory considerations under the 1996 Amendments to the Safe Drinking Water act. These bacteria were place on the CCL occurrence priority list which meant that occurrence information was needed on them. Existing methods for the detection and identification of pathogenic strains for each of these species are mostly laboratory assays and field samples that need to be tested before occurrence studies can be done. In addition, other bacteria such as E. coli 0157:H7, Shigella and Salmonelle spp., which are not on the CCL, also pose substantial risk to human health. Therefore a method that has the potential to detect several bacteria and also identify the pathogenic strains at the same time would be of great value. DNA oligonucleotide microarray offer the potential to simultaneously detect any organism of interest in a water sample and is needed for this study. The following tasks shall be achieved : 1) prepare microarray chips; 2) begin validation of chips with pure cultures; 3) begin validation of chips with spiked and unspiked surface water and 4) deliver chips and prepare report.
