- This is not a notice for a solicitation. This is only a Request for Information (RFI). The Government does not intend to award a contract or any type of agreement on the basis of this RFI or to otherwise pay for the information solicitated under this RFI. This is NOT a request for proposal or an invitation to bid; merely a request for information. The U.S. Geological Survey, National Water Quality Laboratory is considering contracting for the preparation and supply of five microbiology test kits designed to detect and enumerate total coliforms, enterococci, fecal coliforms, Escherichia coli, and fecal streptoccocci in water. These kits are comprised of small bottles containing sterile water and between 2 and 5 powder ingredients, each individually packaged. The ingredients are selective growth agar, indicator dyes, and specific growth media. The kits are aseptically prepared in field offices using 50 x 11 mm Petri plates, pipettes, Erlenmeyer flasks or Griffin beakers, and hot plates. Specific volumes of water samples are filtered through 47 mm membrane filters which are placed on the prepared Petri plates, incubated, and the appropriate bacterial colonies counted and recorded. Each kit contains material sufficient to prepare 100 mL of media and 15 plates.The kits are described below.The bottle sizes, cap sizes, and label colors are required. Total Coliform Medium Kit contains: m ENDO Broth MF - 4.8 g and Bacto Agar - 1.5 g in the same bottle (1 ounce amber glass bottle, red cap, white label), and 2% ethanol in water solution - 100 mL (4 ounce polypropylene bottle, translucent cap, white label) Fecal Coliform Medium Kit contains: m FC Agar - 5.2 g (1 ounce clear glass bottle, white cap, white label), Rosolic acid 0.1 g (1 ounce amber glass bottle, red cap, pink label), 0.2N Sodium Hydroxide Solution - 10 mL (1/2 ounce polypropylene bottle, black cap, pink label), and Reagent Grade Distilled Water - 100 mL (4 ounce polypropylene bottle, translucent cap, white label) Enterococci Basal Medium Kit (EPA Method 1600) contains: mE Agar - 7.12 g (1 ounce clear glass bottle, white cap, white label), Nalidixic acid - 0.025 g (1ounce amber bottle, red cap, white label), 0.2N Sodium Hydroxide Solution - 10 mL (1/2 ounce polypropylene bottle, black cap, pink label), Triphenyltetrazolium Chloride - 0.020 g (1 ounce amber glass bottle, white cap, white label), Reagent Grade Distilled Water - 100 mL (4 ounce polypropylene bottle, translucent cap, white label), Reagent Grade Distilled Water - 10 mL (1/2 ounce polypropylene bottle, black cap, white label), and Indoxyl ?-D-Glucoside - 0.075 g (1 ounce amber glass bottle, red cap, white label) Escherichia coli Substrate Medium Kit (EPA Method 1103) contains: m TEC Agar - 4.53 g (1 ounce clear glass bottle, white cap, white label), Urea - 2.00 g with Phenol Red - 0.01 g (both in same 1 ounce amber glass bottle, red cap, white label), and Reagent Grade Distilled Water - 200 mL in two 100 mL bottles (4 ounce polypropylene bottle, translucent cap, white label) Fecal Streptococcus Medium Kit contains: KF Streptococcus Agar - 7.64 g (1 ounce clear glass bottle, white cap, white label), Reagent Grade Distilled Water - 100 mL (4 ounce polypropylene bottle, Translucent cap, white label), Triphenyltetrazolium Chloride - 0.1 g (1 ounce amber glass bottle, white cap, white label), and Reagent Grade Distilled Water - 10 mL (1/2 ounce polypropylene bottle, black cap, white label) Packaged kits will be quality assured once each month by USGS personnel by randomly selecting one of each type of kit from stock and preparing the Petri dishes as performed in field offices. Three membrane filters are inoculated with pure cultures of bacteria for two positive and one negative test for each kit. These filters are then placed on the appropriate medium in the Petri dishes, incubated, and observed and recorded. These tests indicate the sterility, media selectivity, and colony growth quality of the kits. The pure cultures of bacteria used are Enterococcus faecalis, Escherichia coli, and Enterobacter cloacae. The approximate numbers of kits sold in FY 2003 are: 150 total coliforms, 140 enterococci, 950 fecal coliforms, 375 Escherichia coli, and 300 fecal streptococci. Interested parties that believe they have the expertise and experience to meet this requirement may submit a response to this RFI. The response format should not exceed 10 pages inclusive of facts/figures. Questions and all responses should be submitted to the Contracting Officer, Diana Romero at dlromero@usgs.gov. Responses are due on or before July 30, 2004. Any subsequent solicitation will be synopsized prior to its release.
