- NOTICE OF INTENT TO NEGOTIATE A SOLE SOURCE CONTRACT WITH THE UNIVERSITY OF CALIFORNIA-DAVIS FOR THE NATIONAL MARINE FISHERIES SERVICE (NMFS). PURPOSE OF PROPOSED CONTRACT IS TO ASSESS EFFECTS OF POLLUTANTS IN FISH INJECTED WITH SPMD EXTRACTS. AFTER THE EXXON VALDEZ OIL SPILL, RESEARCH HAS FOCUSED ON THE EXPRESSION OF THE BIOMARKER ENZYME CYP1A IN FISH. THE BIOMARKER CYP1A ACTS AS A MEASURE OF OIL EXPOSURE. PASSIVE SAMPLERS (SEMI-PERMEABLE MEMBRAME DEVICES (SPMD) WILL BE DEPLOYED THROUGHOUT THE OILED AND UN-OILED PARTS OF PRINCE WILLIAM SOUND. EXTRACTS FROM THE SAMPLERS WILL THEN BE ANALYZED TO DETERMINE THEIR POTENTIAL FOR INDUCING CYP1A ENZYMES. NMFS WILL DEPLOY AND COLLECT THE PASSIVE SAMPLERS AND ANALYZE THE EXTRACTS FOR A LARGE NUMBER OF DIFFERENT TYPES OF CONTAMINANTS. THIS STUDY WILL SUPPLEMENT THOSE CHEMICAL MEASUREMENTS WITH ATTEMPTS TO MEASURE THE BIOLOGICAL RESPONSE TO THESE EXTRACTS, TO DETERMINE IF INDUCIBLE POLLUTANTS ARE PRESENT. TO ACCOMPLISH THIS, ALIQUOTS OF CONCENTRATED EXTRACTS FROM PASSIVE SAMPLERS WILL BE INJECTED INTO JUVENILE TROUT, LIVERS WILL BE HARVESTED AT TIMED INTERVALS, AND THE INDUCTION OF CYP1A ACTIVITY WILL BE MEASURED USING EROD ASSAYS. THE PROPOSED CONTRACTOR WILL CONDUCT INJECTIONS AND HARVEST THE LIVERS AND PROVIDE THEM TO ANOTHER CONTRACTOR. AFTER ALL CHEMICAL AND BIOLOGICAL RESPONSE MEASUREMENTS ARE COMPLETED, THE PROPOSED CONTRACTOR WILL EVALUATE AND INTERPRET THE COMBINED RESULTS. METHODS USED TO INDUCE CYP1A ACTIVITY IN FISH USING SPMD EXTRACTS WAS DEVELOPED BY DR. SPRINGMAN AT UC-DAVIS. PREPARATION OF THE EXTRACTS FROM THE SAMPLERS AND THE MEASUREMENT OF EROD ACTIVITY ARE BOTH FAIRLY ROUTINE, BUT NO ONE HAS DEVELOPED THE EXPERTISE TO COMBINE THE TWO METHODS TO DETERMINE AVAILABILITY OF CYP1A STIMULATING POLLUTANTS IN THE ENVIRONMENT. THIS IS A SIMPLIFIED ACQUISITION WITH AN ESTIMATED VALUE OF LESS THAN $100,000.
