Description
The US Department or Agriculture Agricultural Research Service in Hilo, HI has a requirement for a commercially available robot which we will use to conduct large scale fine mapping to define the borders of the male specific region (MSY) of the Y chromosome in papaya, and conduct physical mapping of the papaya genome in the coming year. Fine mapping requires isolation of genomic DNA from thousands of plant leaf samples, while physical mapping requires isolation of DNA from 40,000 bacterial artificial chromosome (BAC) clones. This robot will provide high throughput DNA isolation capacity for both plant and BAC samples, and thus generate more data and make faster progress than isolating DNA manually for the above two and other related projects. Under consideration is a refurbished with full manufactures warranty an Exaction System with the following characteristics Fully automated ?robot? for the isolation of DNA from BAC, Plasmid, and Phage Colonies as well as plant tissues. Capability to provide unattended up to 192 DNA samples in 12-14 hours. Ability to process 0.5-3.0 ml cultures; processing capability of cultures in batches of 24 in specially designed disposable plastic centrifuge tubes units (6 samples per tube unit). Provided with BAC/Plasmid protocol and Plant protocol. Hardware: 6 independent reagent reservoirs delivered by syringe; integral 24 sample swinging bucket rotor centrifuge module; XYZ robotic sample handling; heater module for incubation. Software: menu driven microprocessor control; pre-programmed protocols with limited user programmability; store up to 4 protocols with 7 user files each. Alternate products will certainly be considered which can meet our research requirements as stated above.
