Description
The National Cancer Institute (NCI), Experimental Immunology Branch (EIB), Immune Targeting Section (ITS), intends on entering into a sole source purchase order with Amersham Biosciences, 800 Centennial Avenue, Piscataway, N.J., 08854 for AKTA FPLC w/FRAC 950. The EIB studies the structure and function of molecules used by the innate immune system, particularly Toll-like receptors and related proteins. The ITS is currently expressing TLRs in high amounts for structural analysis. ITS requires an automated chromatography unit that is fully compatible with the Amersham FPLC unit that it replaces and has the ability to purify biomolecules from complex starting materials derived from cell lysates, tissue homogenates, recombinant protein expression systems and other sources. The ITS requires a system with the following capabilities: 1) perform separations based on a variety of sample characteristics, such as molecular weight, net surface charge, hydrophobicity, while retaining biological activity, 2)ability to run large and small columns that contain a wide variety of gel filtrations media, 3) must have a flow rate range of .05-20 ml/min at pressures ranging from 1-700 psi. Because the ITS is using salt buffers, the entire wetted flow patch of the system must be inert to halides, 4) two pump gradient system with dynamic mixing allowing the system to operate with standard low pressure chromatography supports used in affinity chromatography and other techniques, 5) ability to control nine motorized valves used from column selection, buffer selection, flow reversal, and multi-dimensional chromatography, 6) controllers capable of allowing programs to be time-based, volume-based, or column-volume based, 7) equipped with a fraction collector which is fully integrated and controllable via the system control software. Amersham Biosciences is the only source known to the Government that can provide the above features as well as provide a trade-in allowance. However, if any interested party believes it can meet the above requirements, it may submit a statement of capabilities. The capability statement and any other furnished information must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can fully meet the requirements herein. Capability statements must be received in the contracting office no later than 1:30 P.M. EDT on December 8, 2003. This action is pursuant to 41 USC 253 (1) and under the authority of FAR 6.302-1.
