Description
This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in FAR subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. The solicitation number is R69-7MN1-02 and is issued as a request for quotation (RFQ). This solicitation is a small business set-aside; The NAICS is 334516, business size 500 employees. This solicitation document incorporates those provisions and clauses in effect through Federal Acquisition Circular 01-15. The USDA, Agricultural Research Service, requests the procurement of sequencing services for 20,000 ESTs from a normalized cDNA library synthesized from Boophilus microplus. The use of modern molecular approaches for designing novel methods of tick control require increased knowledge in the identification and quantification of expressed genes in the tick and the role specific genes may play in tick development and survival of various stresses encountered by the tick, including dessication, cold, and pesticide exposure. As an initial step in assembling a database of expressed tick genes, a tick cDNA library has been synthesized which contains genes activated by various environmental perturbations and genes expressed in various lifestages of the tick. The objective of this research is to obtain sequence information from genes expressed in the tick with the longer-term goal of relating the expression or activation of specific genes to specific environmental cues. Approach: Expressed sequence tag (EST) data will be compiled into a database of expressed tick genes. Clones from the tick cDNA library will be sequenced from both the 5' and 3' ends using standard sequencing primers. The sequence information will be auto-annotated and putative gene identification assigned. The EST database will be used to build a gene index for Boophilus microplus, assembling sequences into tentative consensus sequences and singletons. Specifications: (1) receive and store the normalized cDNA library; (2) plate the library on selective media, picking bacterial colonies containing cDNA inserts, and preparing DNA for sequencing; (3) Perform 5' and 3' sequencing (20,000 reactions in total); (4) Auto-annotate sequence data, searching for putative open reading frames; (5) Provide sequence information in flat files with putative gene identification when possible; (6) Build a gene index of Boophilus microplus using these ESTs. The Sequencing information shall be delivered to the USDA, Agricultural Research Service, 2700 Fredericksburg Rd, Kerrville, TX 78028. The Government ?REQUIRES? delivery no later than 90 days after receipt of order, however, each offeror shall include their proposed delivery schedule as part of their quotation. Acceptance and FOB point are destination. In order to be considered for award offerors shall provide a quotation on company letterhead detailing the unit pricing; descriptive literature, brochures, and five references for the proposed item.
