Description

The USDA, Agricultural Research Service, intends to enter into a sole source contract with the University of California-Davis, Davis, California. This contract is to procure a library of bacterial artificial chromosome (BAC) clones of the durum wheat variety "Langdon". BAC clones consist of large fragments (greater than 100,000 bp) of DNA cloned into a vector such that they can be maintained and propagated. Large fragment DNA libraries are essential to studies regarding structural and functional genomics including gene isolation, DNA sequencing, DNA marker development, molecular evolutionary studies, comparative genomics, etc. To construct this library, high-molecular weight DNA was obtained from nucei extracted from leaves of "Landon" durum and cloned into the vector pIndigoBAC536. The library consists of 516,096 BAC clones arrayed in 1,344 384-well plates. The library contains less than 1% empty clones and about .34% chloroplast clones. The average insert size of the library is 130,000 bp per clone, and it is estimated to represent 5X durum genome equivalents. It was essentially made by wheat molecular geneticists for use by wheat molecular geneticists. University of California-Davis has the only tetraploid wheat (Langdon) BAC library in the world.
